Spell check will be available in the Visual Editor in 2.5.

For markdown, it's a bit more complicated as the editor itself doesn't support the native browser spellchecking capabilities. A spellcheck mode was added to quickly see misspelt words on the preview panel. The only downside is that the highlighting will disappear as soon as you leave that panel, as focus is required for spellcheck to work.
